Affinity Maturation describes the natural or laboratory-directed process of improving antibody binding affinity through iterative rounds of mutation within complementarity-determining regions and selection for enhanced target engagement, mimicking the somatic hypermutation and clonal selection occurring during natural immune responses in germinal centres.

The biopharmaceutical industry employs in vitro affinity maturation as a standard antibody engineering step improving therapeutic candidate potency following initial discovery. Phage display, yeast display, or ribosome display platforms present mutagenised antibody libraries against target antigens under increasingly stringent conditions, selecting variants with progressively improved affinity. Error-prone PCR, site-directed mutagenesis, and chain shuffling introduce diversity at CDR positions. Deep mutational scanning combined with next-generation sequencing accelerates identification of beneficial mutations. Affinity improvements of 10 to 1000-fold are routinely achieved, enabling lower doses and improved therapeutic indices. As computational design predicts beneficial mutations reducing experimental burden, affinity maturation continues delivering high-potency antibodies for therapeutic development.
