Albumin Binding describes reversible association of drugs, proteins, or nanoparticles with serum albumin, the most abundant plasma protein, profoundly influencing pharmacokinetic behaviour including distribution, half-life extension, and bioavailability. Only unbound drug fractions generally produce pharmacological effects, making albumin binding a critical pharmacokinetic determinant.

The pharmaceutical industry leverages albumin binding both as a challenge requiring characterisation and as a deliberate half-life extension strategy. Lipidated peptides like semaglutide intentionally exploit albumin binding enabling once-weekly dosing. Albumin-drug conjugates deliver cytotoxics exploiting tumour accumulation. Analytical methods including equilibrium dialysis and ultrafiltration measure binding extent. High albumin binding may reduce free drug concentrations affecting dose requirements. Displacement interactions between co-administered highly bound drugs may significantly alter free drug concentrations requiring clinical monitoring and dosing guidance in drug labels.
