Allometric Scaling

Allometric scaling predicts human pharmacokinetic or physiological parameters from animal data using mathematical relationships based on body size and species differences.

Allometric Scaling designates the mathematical approach using power law relationships between body weight and physiological parameters to predict pharmacokinetic properties across species, enabling translation of animal data to human dose predictions during drug development. This empirical methodology accounts for species differences in metabolic rates, organ sizes, and physiological function.

The pharmaceutical industry applies allometric scaling during IND-enabling programmes to predict human clearance, volume of distribution, and half-life from preclinical species data, informing first-in-human dose selection with appropriate safety margins. Simple allometry, multi-species scaling, and physiologically based pharmacokinetic modelling represent increasingly sophisticated predictive approaches. Brain weight correction may improve predictions for centrally acting drugs. Validation demonstrates reasonable predictive accuracy for many compound classes. As predictive modelling advances through expanded databases and machine learning, allometric scaling continues providing practical human dose prediction frameworks supporting early clinical development.
