Basophil Activation

Basophil Activation describes the stimulation of basophils, rare circulating granulocytes expressing high-affinity IgE receptors, leading to rapid degranulation and release of histamine, cytokines, and other inflammatory mediators upon allergen exposure or immune stimulus. This activation contributes to allergic responses, anaphylaxis, and certain immune regulatory functions.

The pharmaceutical industry studies basophil activation as a biomarker for allergy diagnosis, drug hypersensitivity assessment, and evaluation of anti-allergic therapeutics. The basophil activation test using flow cytometry measures CD63 or CD203c upregulation following allergen challenge, providing functional evidence of IgE-mediated sensitisation. Monoclonal antibodies targeting IgE including omalizumab reduce basophil sensitivity by depleting free IgE, demonstrating how basophil biology informs therapeutic targeting. Safety assessment for new biologics includes monitoring for basophil-mediated hypersensitivity signals in clinical trials.
