Buffer Exchange

Buffer exchange replaces one buffer system with another while retaining the target biomolecule, typically to support purification, formulation or analytical testing.

Buffer Exchange designates the process of replacing the buffer system surrounding macromolecules or particles in solution with a different buffer composition while retaining the target molecule, commonly accomplished through diafiltration using ultrafiltration membranes, size-exclusion chromatography, or dialysis. This operation is critical for preparing biologics for formulation or further processing steps.

The biopharmaceutical industry employs buffer exchange throughout downstream processing and formulation development. Diafiltration using tangential flow filtration systems efficiently exchanges buffers at large scale while concentrating products simultaneously, making it the preferred method for manufacturing operations. The process removes process-related impurities including residual chromatography buffers, aggregation-prone co-solvents, or excipients incompatible with subsequent steps.
